Drug Title

Uroxatral (Alfuzosin)

Pharmacological action

An alpha-adrenoblocker acting predominantly on postsynaptic α1-adrenoreceptors. By blocking α1-adrenoreceptors located in the triangle and sphincter of the bladder and urethra, alfuzosin eliminates spasm of smooth muscle fibers, which leads to a decrease in resistance of outflow of urine. It reduces the tone of smooth muscle of arteries and veins. Reduces OPPS and systemic BP.

Pharmacokinetics

After oral administration, alfuzosin is absorbed from the gastrointestinal tract. Cmax in plasma is reached after 3 hours and is 10.3 ng/ml. Differences in maximum and minimum plasma concentrations are insignificant. Food intake has no effect on absorption.


Binding to plasma proteins is about 90%. Metabolized in the liver. T1/2 is 8 hours. 15-30% of inactive metabolites are excreted in the urine, 75-91% - in the faeces. 11% of alfuzosin is excreted unchanged in the urine.


In patients over 75 years old a higher plasma concentration and bioavailability of alfuzosin is noted (possibly due to reduced metabolism in the liver). In patients with significant renal impairment, alfuzosin clearance is increased (possibly due to less binding to blood plasma proteins).

Side effects

Digestive system: epigastric pain, nausea, diarrhea, rarely - dry mouth.


CNS: dizziness, headache, rarely - drowsiness.


Cardiovascular system: rare - tachycardia, orthostatic hypotension.


Allergic reactions: rare - skin rash and itching.


Other: malaise, rarely - edema, pain behind the chest.


Contraindications to use

Indications in the anamnesis of orthostatic hypotension, concomitant use of other alpha-adrenoblockers, hypersensitivity to alfuzosin.

Administration during pregnancy and lactation

The drug is not intended for use in women.


Administration in patients with kidney dysfunction

Caution is required when using in patients with significant renal dysfunction.

Administration in children

The drug is not intended to be used in children.

Administration in elderly patients

For patients older than 65 years old - the initial dose is 2.5 mg 2 times per day (in the morning and in the evening); further the dose can be increased (but no more than 10 mg/day).


Use with caution in patients over 75 years of age.

Indications of active substances of the drug Alfuzosin

Treatment of functional manifestations of benign prostatic hyperplasia when surgical intervention is not possible, as well as in cases of progressive growth, especially in elderly patients.
